Feedback inhibition of aspartate transcarbamylase in liver and in hepatoma.

چکیده
Aspartate transcarbamylase was partially purified from rat liver, Morris hepatoma 5123-A or D, Hepatoma 7800, and Novikoff hepatoma and was shown to be inhibited by pyrimidine deoxyribonucleosides and deoxyribonucleotides. The pyrimidine ana logs, 5-bromo-2'-deoxyuridine, 5-bromouridine, 5-bromo-2'-deoxycytidine, 5-fluoro2'-deoxyuridine, 5-fluorouridine, 5-fluoro-2'-deoxycytidine, 5-iodo-2'-deoxyuridine, al

Carbamyl phosphate-aspartate transcarbamylase activity in regenerating rat liver.
An increase in CP-asp. 1 transcarbamylase activity in experimentally produced rat hepatoma was reported in a previous communication (5). The question whether the increased activity is characteristic of neoplasia or is simply a reflection of the metabolic changes taking place in rapidly growing tissue presented itself. Since liver tissue has the capacity to grow relatively rapidly during the pro...

SUMMARY Qualitative and quantitative studies on crude preparations of aspartate transcarbamylase from normal and neoplastic human colon were carried out. Whereas the activity of the en zyme was markedly higher in neoplastic tissue, no qualitative differences were observed between the enzymes from the two types of tissues. Negative feedback inhibition of human colon aspartate transcarbamylase co...
متن کاملCarbamyl phosphate-aspartate transcarbamylase activity in tumors.
The enzyme carbamyl phosphate-aspartate transcarbamylase catalyzes the transfer of the carbamyl group from carbamyl phosphate (CP) to L-aspartate ($). Preliminary experiments with a series of six relatively advanced azo dye-induced hepatomas indicated that hepatoma tissue had a higher CP-aspartate transcarbamylase activity than did liver from normal animals. Experiments are reported here in whi...
متن کاملElimination of cooperativity in aspartate transcarbamylase by nitration of a single tyrosine residue.
In a previous report [Landfear, S. M., Lipscomb, W. N. & Evans, D.R. (1978) J. Biol. Chem. 253, 3988--3996] we demonstrated that tetranitromethane can be employed to nitrate a limited number of tyrosine residues in aspartate transcarbamylase (carbamoylphosphate:L-aspartate carbamoyltransferase, EC 2.1.3.2); such modification eliminates cooperativity, feedback inhibition, and enzymatic activity,...
متن کاملPyrimidine biosynthesis de novo in M. leprae.
Mycobacterium leprae can synthesise pyrimidines de novo. Although pyrimidine synthesis could not be detected in intact bacteria, extracts contained all four enzymes unique to the de novo pathway which are detectable in mycobacteria by the methods used. Inhibition of aspartate transcarbamylase by UTP and ATP suggested that lack of pyrimidine synthetic activity in whole M. leprae could be a resul...
